Abstract

Thin layer chromatographic purity determination of commercial desipramine hydrochloride (active ingredient of Pertofran®) and clomipramine hydrochloride (active ingredient of Anafranil®) shows the presence of some impurities, the identity and quantity of which are investigated. With the exception of imipramine and dichloroimipramine none of the contaminants exceeds 0,1%. For desipramine the artifact nature of a TLC side spot is demonstrated and elucidated. One of the trace contaminants (4-methyl-desipramine) found in desipramine originates from an analogous and also identified compound in imipramine hydrochloride (active ingredient of Tofranil®). Pharmacological studies give no indications for a possible cause of unfavourable side effects by these trace impurities.
